ASSAY PROCEDURE
all standards, control, and samples 모두 duplicate.
사용전에 RT 꺼내 두기
1. 사용할 plate frame 제외하고 나머지 다시 밀봉해서 넣어둠
2. RD1N 를 사용 중에 잘 흔들어 준후 50 μL 씩 각 well에 넣어줌
3. 각웰에 50 μL of standard, control, or sample 넣고. 뚜껑 닫은 후 가볍게 1분 정도 섞어줌. 상온에서 2h.
4. 2시간후 Wash Buffer (400 μL) *5회 세척
5. 100 μL of Mouse Mouse IL-1β Conjugate 넣고 새로운 뚜껑 덮어준 후 RT에서 2시간
6. 2시간후 Wash Buffer (400 μL) *5회 세척
7. Add 100 μL of Substrate Solution , 빛 가린 후 30분 RT
8. 100 μL of Stop Solution.
9. 30분이내 set to 450 nm. readings
(If wavelength correction is available, set to 540 nm or 570 nm.
If wavelength correction is not available, subtract readings at 540 nm or 570 nm from the readings at 450 nm.)
